It will be the funny man's first appearance on the Oscars stage.
The 38-year-old said of his selection, "I will do my utmost to live up to the high standards set forth by my predecessors… and I hope they don’t find out I hosted the Charlie Sheen Roast."
Oscar telecast producer Neil Meron called MacFarlane "the most unbelievable, consummate host choice we could think of".
MacFarlane's film Ted is one of 2012's highest box office grossers, bringing in over $420 million worldwide.
The 85th Annual Academy Awards will take place on Sunday February 24.
